This report describes the on-site commissioning of a peptide synthesizer at a pharmaceutical research institute and its validation through a complete solid-phase synthesis of thymopentin (TP-5). The peptide synthesizer was prepared through system software upgrade, reagent-line cleaning, flow-rate calibration, and method optimization, then verified with a full TP-5 run. Results confirmed stable operation and accurate reagent delivery suitable for routine peptide production.

When a customer asks whether our peptide synthesizer can handle real production, the best answer is to run it. Our engineer went to a pharmaceutical research institute, set up the instrument, and synthesized thymopentin (TP-5) from scratch. A peptide synthesizer is only as good as the batch it actually delivers, so we treat every install like a live test. What we checked before the first run

Upgraded the control software to the latest stable build on the peptide synthesizer.
Flushed every line with methanol and NMP, three passes each, so the peptide synthesizer starts clean.

One problem we fixed
Resin was washing to the top of the reactor and throwing off the flow meter of the peptide synthesizer. A parameter tweak in the program solved it on the peptide synthesizer. Small fix, big impact on accuracy for any peptide synthesizer doing SPPS.
